Melanin formation inhibition test was conducted using B16 melanoma cells

 

 

 1 x 10 ⁵ B16 melanoma cells were planted in a 60 mm plastic culture dish and were cultured 24 hours earlier. Then they were transferred to a fresh culture medium and test materials were added to the culture medium, each at a concentration of 2%.

 Two days after the addition of these materials, the cells were recovered by processing with trypsin and dissolved with heat in a 10% DMSO solution; then the absorbance was measured at 475 nm. At the same time the activity of the cells, which were supplemented with various materials, was measured by the MTT reduction method. The MTT reduction activity of the loaded cells in the various trials was calculated, considering the MTT reduction activity as one unit of the control cell.

 Calculating the quantity of melanin formed in the various trials in terms of units of absorbance/MTT reduction activity, the rate of inhibition was sought for the amount of melanin formed in the control cells.

Culture medium : Eagle MEM culture medium including 10% FBS  

 

Cell proliferation test is conducted using human fibroblast cells

 

W e used normal human dermal fibroblast cells (NB1RGB) and cultured them in MEM culture medium including 5% FBS. It was seeded on a 96 well micro plate, 5 x 103 cell/well, to get the right density.

 24 hours later, various materials were exchanged and supplemented in the medium in order to produce the right concentrations. At this point a control was prepared with certain materials omitted.

 While exchanging the materials every 3 days, the culture was continued in a CO2 incubator for 9 days, and then the cell reproduction rate was measured by the MTT reduction method. 

  MEM culture medium including 5% FBS : Then the FBS concentration that would neither increase nor reduce the number of cells was studied.
